We are proud to announce the manuscript preprint for our TOBIAS software, which is the first comprehensive tool for footprinting analysis on ATAC-seq.

In this work we present TOBIAS, which is as a comprehensive framework for footprinting analysis. TOBIAS is the first of its kind to integrate both preprocessing, footprinting, visualization and downstream analysis in a single framework. As a proof of concept we used TOBIAS to investigate the kinestics of transcription factor binding during zygotic genome activation (ZGA). We show that by using the Tn5-bias correction algorithm of TOBIAS, it is possible to visualize protein binding with gain and loss of footprints throughout development. We also used these data to draw parallels between human and mouse developmental stages and highlight transcription factors which have diverged in function. By going into more detail with the ZGA-regulator Dux, we further highlight the capabilities of TOBIAS to predict specific target lists, predict TF-activation networks and uncover novel genetic elements.
